● 產品編號及規(gui)格:

本(ben)產品是由6條帶狀雙鏈DNA條帶組成(cheng),適用(yong)(yong)于瓊脂糖凝膠電(dian)(dian)泳(yong)中(zhong)DNA條帶的分(fen)析。產品中(zhong)已含有1×loading buffer,可(ke)根據(ju)實驗(yan)需要,直接取(qu)2-5μl電(dian)(dian)泳(yong),使用(yong)(yong)方(fang)便(bian),電(dian)(dian)泳(yong)圖像清晰。6條帶的大小和含量(liang)分(fen)別為100(50 ng/5 μl),200(50 ng/μl),300(50 ng/μl),400100 ng/μl,500(50 ng/μl),600(50 ng/μl) bp。其中400bp條帶(dai)加(jia)亮顯示。

● 儲存及運輸:

 4℃貯存(cun)3個月, -20℃長期貯存(cun);常(chang)溫運輸。



● 使(shi)用方(fang)法:

1. 取2-5 μl本產品加入到瓊(qiong)脂糖(tang)凝膠的加樣孔中就行(xing)電泳(yong)。

注(zhu):根據電泳梳子的厚(hou)度(du)和寬度(du)確定上樣(yang)量。每(mei)1mm×1mm(厚度×寬(kuan)度)加樣孔上樣1μl;窄齒(chi)梳(shu)子(一(yi)般(ban)為1mm×2mm)上樣2μl;寬(kuan)齒梳子(一(yi)般為1mm×5mm)上(shang)樣5μl。如(ru)果使用厚齒梳子或寬于5mm的梳子,可(ke)以適當(dang)調整上樣量。

2. 建議電泳條件:

凝膠濃度(du)為(wei)2.0%,凝膠長度5-7cm,電泳電壓(ya)~7v/cm,電泳時間30-35分鐘。

3. 通過核酸染料染色后紫外燈或藍光儀下觀察(cha)條帶。

● 注意事項:

1. 瓊脂(zhi)糖的(de)(de)質量(liang)對DNA的(de)(de)電(dian)泳(yong)有很大(da)影(ying)響(xiang),電(dian)泳(yong)時(shi)請盡量(liang)使用(yong)(yong)質量(liang)優等的(de)(de)瓊脂(zhi)糖。由于PAGE電(dian)泳(yong)的(de)(de)特殊性,50 bp DNA ladder 不建議用(yong)(yong)于PAGE電(dian)泳(yong)。

2. 使用(yong)與電泳緩(huan)(huan)沖液一(yi)致的緩(huan)(huan)沖液融化瓊(qiong)脂(zhi)糖,例(li)如使用(yong)1×TAE緩(huan)(huan)沖液電泳,需(xu)使用(yong)1×TAE緩(huan)(huan)沖液溶膠(jiao)。

3. 多次電(dian)泳后緩沖液電(dian)導增(zeng)強,相同電(dian)壓下電(dian)流增(zeng)大,導致分辨率降低,并且(qie)明顯產(chan)熱(re),嚴重(zhong)時會引起(qi)凝膠熔化(hua)或DNA變性。

4. 推薦使用RealSafe系列染(ran)料(liao)(liao)(貨號(hao):GR002,GR003,GR004)染(ran)色。如果使用Goldview或EB等染(ran)料(liao)(liao)進行染(ran)色,由于此類染(ran)料(liao)(liao)帶更多的(de)(de)正(zheng)電(dian)荷(染(ran)料(liao)(liao)移(yi)動方(fang)向與核(he)酸泳(yong)(yong)動方(fang)向相反),隨著電(dian)泳(yong)(yong)時間(jian)延長(chang),染(ran)料(liao)(liao)向大(da)片段聚集,凝膠會(hui)出現陰陽(yang)(yang)膠現象(染(ran)料(liao)(liao)含(han)量(liang)(liang)高的(de)(de)凝膠紫(zi)(zi)外下較亮,含(han)量(liang)(liang)低(di)的(de)(de)凝膠紫(zi)(zi)外下發暗),導(dao)致小分(fen)(fen)子(zi)(zi)量(liang)(liang)DNA顯色很弱(ruo)或不可見(jian)。陰陽(yang)(yang)膠可以(yi)在(zai)電(dian)泳(yong)(yong)結束后浸泡染(ran)色,將膠浸沒在(zai)含(han)1 μg /ml EB或Goldview的(de)(de)電(dian)泳(yong)(yong)緩沖(chong)液中20-25 min,小分(fen)(fen)子(zi)(zi)量(liang)(liang)DNA會(hui)顯色清楚。更長(chang)時間(jian)浸泡會(hui)因DNA分(fen)(fen)子(zi)(zi)擴散而使條帶彌(mi)散,小分(fen)(fen)子(zi)(zi)量(liang)(liang)DNA更易(yi)彌(mi)散。

5. 紫外照(zhao)射下(xia)EB易分解(jie),使(shi)DNA條帶熒光變淺(qian)。DNA受紫外光照(zhao)射形成嘧啶二聚體,不利于(yu)待(dai)回收片段的下(xia)游(you)工作,因此盡可能減少紫外照(zhao)射時間。

6. 蔗糖(tang)會(hui)結合DNA,降低(di)其電泳(yong)遷移率,建議DNA樣(yang)(yang)品上樣(yang)(yang)使用(yong)含甘油的Loading Buffer。

7. 大部(bu)分核(he)酸工(gong)具酶會(hui)結(jie)合DNA,降(jiang)低其電泳遷移率,大分子量DNA尤為明顯。
