本(ben)產品(pin)是(shi)由11條帶狀雙(shuang)鏈DNA組(zu)成,適用于瓊(qiong)脂糖凝膠電(dian)泳(yong)中DNA條帶(dai)的分析。本產品為(wei)即用型產品,已含有(you)1×loading buffer,可根據實驗需(xu)要,直(zhi)接(jie)取(qu)2-5μl電泳,使(shi)用方便,電泳圖像清晰。 本(ben)產品中11條帶為10020030040050060070080090010001500bp,其中500bp條帶(dai)為100ng/5μl,其余條帶濃(nong)度(du)為(wei)50ng/5μl


● 儲存條件:

  4 貯存6個月(yue),-20貯存 1年。

● 使用方法:

1. 取(qu)2-5 μl本產品(pin)加(jia)入到瓊(qiong)脂糖凝膠的加(jia)樣孔中(每1mm×1mm加(jia)樣孔加(jia)1μl,如果加(jia)樣孔寬于(yu) 

5mm,可以適(shi)當增加上樣量)進(jin)行電泳。

2. 建議電泳條件:凝膠濃度為1.0%,凝膠長度10 cm,電泳電壓~7 v/cm,電泳時間50分鐘+

3. 通(tong)過(guo)核酸染料染色后紫(zi)外燈或藍光儀下觀察(cha)條帶。

● 注意事項:

1. 瓊脂糖(tang)的(de)(de)質量對DNA的(de)(de)電(dian)泳(yong)(yong)有很大影響,電(dian)泳(yong)(yong)時請盡量使用質量優等的(de)(de)瓊脂糖(tang)。由于(yu)PAGE電(dian)泳(yong)(yong)的(de)(de)特殊性,該產品不建議用于(yu)PAGE電(dian)泳(yong)(yong)。

2. 使用(yong)與電泳(yong)緩沖(chong)液(ye)(ye)(ye)一致(zhi)的緩沖(chong)液(ye)(ye)(ye)融化瓊脂(zhi)糖,例(li)如使用(yong)1×TAE緩沖(chong)液(ye)(ye)(ye)電泳(yong),需使用(yong)1×TAE緩沖(chong)液(ye)(ye)(ye)溶(rong)膠。

3. 多次電(dian)(dian)(dian)泳后緩(huan)沖液(ye)電(dian)(dian)(dian)導增(zeng)強(qiang),相同(tong)電(dian)(dian)(dian)壓下電(dian)(dian)(dian)流增(zeng)大,導致分辨率降低,并且(qie)明顯(xian)產熱,嚴重(zhong)時會(hui)引起(qi)凝膠熔(rong)化或DNA變性。

4. 推薦(jian)使(shi)用RealSafe系列染(ran)(ran)料(liao)(liao)(貨(huo)號:GR002,GR003,GR004)染(ran)(ran)色(se)。如果使(shi)用Goldview或(huo)EB等染(ran)(ran)料(liao)(liao)進行染(ran)(ran)色(se),由于(yu)此類染(ran)(ran)料(liao)(liao)帶更多的(de)正電(dian)荷(染(ran)(ran)料(liao)(liao)移動方向(xiang)(xiang)與核酸泳(yong)動方向(xiang)(xiang)相反),隨著(zhu)電(dian)泳(yong)時(shi)間(jian)延長(chang),染(ran)(ran)料(liao)(liao)向(xiang)(xiang)大片段聚集,凝(ning)(ning)膠會出現陰陽膠現象(染(ran)(ran)料(liao)(liao)含(han)量(liang)高的(de)凝(ning)(ning)膠紫外下(xia)(xia)較亮,含(han)量(liang)低的(de)凝(ning)(ning)膠紫外下(xia)(xia)發暗),導致小分子(zi)量(liang)DNA顯(xian)色(se)很弱或(huo)不可(ke)見。陰陽膠可(ke)以在電(dian)泳(yong)結束(shu)后(hou)浸泡染(ran)(ran)色(se),將膠浸沒在含(han)1 μg /ml EB或(huo)Goldview的(de)電(dian)泳(yong)緩(huan)沖(chong)液中20-25 min,小分子(zi)量(liang)DNA會顯(xian)色(se)清楚。更長(chang)時(shi)間(jian)浸泡會因DNA分子(zi)擴(kuo)散而使(shi)條帶彌散,小分子(zi)量(liang)DNA更易彌散。

5. 紫(zi)外(wai)照射(she)下EB易分解,使DNA條帶熒光變(bian)淺。DNA受紫(zi)外(wai)光照射(she)形成嘧(mi)啶二(er)聚體(ti),不(bu)利于(yu)待回收片段的下游工作,因此盡可能減少(shao)紫(zi)外(wai)照射(she)時間。

6. 蔗糖會結合DNA,降低其電泳遷移率,建(jian)議DNA樣品上樣使用含甘油的Loading Buffer。

7. 大部(bu)分核酸工具酶會結合DNA,降低其(qi)電泳(yong)遷移率(lv),大分子量DNA尤為明(ming)顯。
