    本(ben)產品是由(you)8條帶狀雙(shuang)鏈DNA條帶組成,適用于瓊脂糖凝膠電泳中DNA條帶的分析。

本(ben)產品為(wei)即用型產品,已含有1×loading buffer,可根據實驗(yan)需要,直(zhi)接取2-5μl電泳,使用方便,電泳圖像清(qing)晰。本(ben)產品中8條帶分(fen)為(wei)1000(50 ng/5 μl),2000(50 ng/5 μl),3000(50 ng/5 μl),4000(100 ng/5 μl),5000(50 ng/5 μl),6000(50 ng/5 μl),8000(50 ng/5 μl),10000bp(50 ng/5 μl)。其中4000bp條帶加亮顯示。


● 儲存及運輸:

4貯存3個月, -20長(chang)期貯存;常溫運輸(shu)。

● 使用方法:

1. 取2-5μl本產品(pin)加(jia)入到瓊(qiong)脂糖凝(ning)膠(jiao)的加(jia)樣孔中(zhong)(每1mm×1mm加樣孔(kong)加1μl,如(ru)果加樣孔(kong)寬于5mm,可(ke)以(yi)適當(dang)增加上(shang)樣量)就行電泳。

2. 建(jian)議電泳條件:凝膠濃度為0.8-1.0%,凝膠長度10 cm,電泳電壓~7v/cm,電(dian)泳(yong)時間60+分鐘。

3. 通(tong)過核(he)酸染料染色后紫外燈或藍光儀下觀察條帶。


● 注意事(shi)項:

1. 瓊脂糖的(de)質(zhi)量對DNA的(de)電泳(yong)(yong)有很大影響,電泳(yong)(yong)時請盡(jin)量使用(yong)質(zhi)量優等的(de)瓊脂糖。由于PAGE電泳(yong)(yong)的(de)特殊性(xing),該(gai)產品不建議用(yong)于PAGE電泳(yong)(yong)。

2. 使用(yong)與電泳緩(huan)沖(chong)液(ye)(ye)一致的緩(huan)沖(chong)液(ye)(ye)融化瓊脂(zhi)糖,例如使用(yong)1×TAE緩(huan)沖(chong)液(ye)(ye)電泳,需使用(yong)1×TAE緩(huan)沖(chong)液(ye)(ye)溶膠(jiao)。

3. 多(duo)次(ci)電泳后緩沖液電導增(zeng)強,相同電壓下電流增(zeng)大,導致分(fen)辨率降低,并且明顯產熱,嚴重時會引(yin)起凝膠熔(rong)化(hua)或DNA變性。

4. 推薦使用RealSafe系列染(ran)(ran)料(liao)(貨號(hao):GR002,GR003,GR004)染(ran)(ran)色(se)。如果使用Goldview或EB等(deng)染(ran)(ran)料(liao)進行染(ran)(ran)色(se),由于(yu)此類染(ran)(ran)料(liao)帶(dai)更(geng)多(duo)的(de)正電荷(染(ran)(ran)料(liao)移動方(fang)向與(yu)核酸泳動方(fang)向相反),隨著(zhu)電泳時(shi)間(jian)延長,染(ran)(ran)料(liao)向大片段(duan)聚集,凝膠(jiao)會(hui)出現(xian)陰陽(yang)(yang)膠(jiao)現(xian)象(染(ran)(ran)料(liao)含(han)量(liang)高的(de)凝膠(jiao)紫外下較(jiao)亮,含(han)量(liang)低的(de)凝膠(jiao)紫外下發暗(an)),導致(zhi)小(xiao)分(fen)(fen)子(zi)量(liang)DNA顯色(se)很(hen)弱或不可(ke)見。陰陽(yang)(yang)膠(jiao)可(ke)以在(zai)電泳結束后浸泡染(ran)(ran)色(se),將(jiang)膠(jiao)浸沒在(zai)含(han)1 μg /ml EB或Goldview的(de)電泳緩沖液中20-25 min,小(xiao)分(fen)(fen)子(zi)量(liang)DNA會(hui)顯色(se)清楚。更(geng)長時(shi)間(jian)浸泡會(hui)因(yin)DNA分(fen)(fen)子(zi)擴(kuo)散(san)(san)而使條帶(dai)彌散(san)(san),小(xiao)分(fen)(fen)子(zi)量(liang)DNA更(geng)易彌散(san)(san)。

5. 紫外(wai)照(zhao)射下EB易分解(jie),使DNA條帶(dai)熒光變淺。DNA受紫外(wai)光照(zhao)射形成(cheng)嘧啶二聚體,不利于待回收片段的下游工作,因此盡可能(neng)減少紫外(wai)照(zhao)射時間(jian)。

6. 蔗糖會(hui)結合DNA,降低其電泳遷移(yi)率(lv),建議DNA樣品上(shang)樣使(shi)用含甘油的Loading Buffer。

7. 大(da)部分(fen)核酸(suan)工具(ju)酶會(hui)結合DNA,降低其電泳遷移率,大(da)分(fen)子(zi)量DNA尤為明顯。
