● 產品編號及(ji)規格:

        RTM436-02       100T(2×250μl)

本產品是(shi)由7條(tiao)(tiao)帶狀(zhuang)雙鏈DNA條(tiao)(tiao)帶組成,適用于(yu)瓊(qiong)脂糖凝膠電(dian)泳中DNA條(tiao)(tiao)帶的(de)分(fen)析。產品中已含有1×loading buffer,可根(gen)據實驗需要,直接取2-5 μl電(dian)泳,使用方便(bian),電(dian)泳圖(tu)像(xiang)清晰。7條(tiao)(tiao)帶的(de)大小和含量分(fen)別為100(50 ng/5 μl),200(50 ng/μl),300(50 ng/μl),400100 ng/μl,500(50 ng/μl),600(50 ng/μl),700(50 ng/μl)bp。其中400bp條帶加亮顯示。


● 儲存及運輸:

   4貯(zhu)存3個月, -20長(chang)期(qi)貯(zhu)存;常溫運輸。

● 使用方法:

1. 取2-5 μl本產品加入到(dao)瓊脂糖凝膠的加樣孔中就(jiu)行(xing)電泳(yong)。

注:根據(ju)電泳(yong)梳(shu)子(zi)的厚度(du)和寬度(du)確(que)定上(shang)樣量。每1mm×1mm(厚度×寬(kuan)度)加樣孔上樣1μl;窄齒梳子(一般(ban)為1mm×2mm)上(shang)樣2μl;寬齒梳子(一般為1mm×5mm)上樣5μl。如果使用厚齒(chi)梳(shu)子或(huo)寬于(yu)5mm的梳子,可(ke)以(yi)適(shi)當調整上樣量。

2. 建議電(dian)泳條(tiao)件(jian):凝膠濃度為2.0%,凝膠長(chang)度5-7 cm,電(dian)(dian)泳(yong)電(dian)(dian)壓(ya)~7 v/cm,電(dian)泳時間30-35分鐘。

3. 通過核酸染料染色后紫外燈(deng)或藍光儀下(xia)觀察條(tiao)帶(dai)。

● 注意(yi)事項:

1. 瓊脂糖的質量(liang)對(dui)DNA的電(dian)(dian)泳(yong)有很大影(ying)響(xiang),電(dian)(dian)泳(yong)時請盡量(liang)使(shi)用質量(liang)優(you)等的瓊脂糖。由于PAGE電(dian)(dian)泳(yong)的特殊(shu)性,該產(chan)品(pin)不建議(yi)用于PAGE電(dian)(dian)泳(yong)。

2. 使(shi)用(yong)與電泳緩沖(chong)液一致的緩沖(chong)液融化瓊脂(zhi)糖,例如使(shi)用(yong)1×TAE緩沖液電泳,需(xu)使(shi)用(yong)1×TAE緩沖液溶膠。

3. 多次電(dian)泳后緩沖(chong)液電(dian)導(dao)增強,相同(tong)電(dian)壓下電(dian)流增大,導(dao)致分辨率降低(di),并(bing)且明顯(xian)產熱,嚴重(zhong)時會引(yin)起凝膠熔化或(huo)DNA變性。

4. 推(tui)薦使用RealSafe系列染料(liao)(貨號:GR002,GR003,GR004)染色(se)。如果使用Goldview或(huo)EB等染料(liao)進(jin)行(xing)染色(se),由于此類染料(liao)帶更(geng)(geng)多的(de)(de)正電(dian)(dian)荷(染料(liao)移動方向(xiang)與核酸泳(yong)動方向(xiang)相反(fan)),隨著(zhu)電(dian)(dian)泳(yong)時間延長(chang),染料(liao)向(xiang)大(da)片(pian)段聚(ju)集,凝(ning)膠(jiao)會出現陰陽膠(jiao)現象(染料(liao)含量高的(de)(de)凝(ning)膠(jiao)紫外下較亮,含量低的(de)(de)凝(ning)膠(jiao)紫外下發暗),導致(zhi)小分子(zi)量DNA顯(xian)色(se)很(hen)弱(ruo)或(huo)不可(ke)見(jian)。陰陽膠(jiao)可(ke)以(yi)在電(dian)(dian)泳(yong)結(jie)束后浸(jin)泡染色(se),將膠(jiao)浸(jin)沒在含1 μg /ml EB或(huo)Goldview的(de)(de)電(dian)(dian)泳(yong)緩沖液中(zhong)20-25 min,小分子(zi)量DNA會顯(xian)色(se)清(qing)楚。更(geng)(geng)長(chang)時間浸(jin)泡會因DNA分子(zi)擴散而使條帶彌(mi)散,小分子(zi)量DNA更(geng)(geng)易(yi)彌(mi)散。

5. 紫(zi)外(wai)照(zhao)射(she)下EB易分(fen)解,使DNA條帶熒光(guang)變淺。DNA受紫(zi)外(wai)光(guang)照(zhao)射(she)形成嘧啶二聚體,不利(li)于(yu)待回收片段的(de)下游工作,因(yin)此盡可能減少(shao)紫(zi)外(wai)照(zhao)射(she)時間(jian)。

6.  蔗(zhe)糖會結合(he)DNA,降低其電泳遷移率,建議DNA樣品上樣使用含甘油的Loading Buffer。

7. 大(da)部(bu)分核(he)酸工(gong)具酶會結合DNA,降低其電泳遷移率,大(da)分子量(liang)DNA尤為明顯(xian)。
