● 產品簡介(jie):

    本產品(pin)是由8條帶(dai)狀雙(shuang)鏈DNA條帶(dai)組成,適用(yong)于(yu)瓊脂糖凝膠電泳中DNA條帶(dai)的分析。

本產(chan)品為即用(yong)型產(chan)品,已含有(you)1×loading buffer,可根據(ju)實驗需要,直接取2-5μl電泳,使用(yong)方便,電泳圖像清晰。本產(chan)品中8條帶分為1000(50 ng/5 μl),2000(50 ng/5 μl),3000(50 ng/5 μl),4000(100 ng/5 μl),5000(50 ng/5 μl),6000(50 ng/5 μl),8000(50 ng/5 μl),10000bp(50 ng/5 μl)。其中4000bp條帶加亮(liang)顯示。


● 儲存及運(yun)輸:

4貯存3個月, -20長期貯(zhu)存;常溫(wen)運輸。

● 使(shi)用方法:

1. 取2-5μl本(ben)產品加入到瓊(qiong)脂糖(tang)凝膠(jiao)的加樣孔中(每1mm×1mm加(jia)樣(yang)孔加(jia)1μl,如果加(jia)樣(yang)孔寬于5mm,可以適當增加上(shang)樣(yang)量(liang))就行電泳。

2. 建(jian)議電泳條件(jian):凝膠濃(nong)度為0.8-1.0%,凝膠長(chang)度10 cm,電(dian)泳電(dian)壓(ya)~7v/cm,電泳時間60+分鐘。

3. 通過核酸染(ran)料染(ran)色后紫外(wai)燈(deng)或藍光儀(yi)下觀察條(tiao)帶(dai)。


● 注意事項:

1. 瓊脂糖的(de)(de)質量對(dui)DNA的(de)(de)電泳有很(hen)大影響(xiang),電泳時(shi)請盡量使用質量優(you)等的(de)(de)瓊脂糖。由于PAGE電泳的(de)(de)特殊性,該產品不(bu)建議用于PAGE電泳。

2. 使用(yong)與電泳緩沖(chong)液一致的緩沖(chong)液融化瓊(qiong)脂糖,例如(ru)使用(yong)1×TAE緩沖(chong)液電泳,需使用(yong)1×TAE緩沖(chong)液溶膠。

3. 多次電(dian)(dian)泳后緩沖液電(dian)(dian)導(dao)增(zeng)強,相同電(dian)(dian)壓(ya)下電(dian)(dian)流(liu)增(zeng)大,導(dao)致分辨率降(jiang)低,并且明顯產(chan)熱(re),嚴(yan)重時會引(yin)起凝膠熔化(hua)或DNA變(bian)性(xing)。

4. 推薦使(shi)(shi)用RealSafe系列染(ran)(ran)(ran)(ran)(ran)料(貨(huo)號:GR002,GR003,GR004)染(ran)(ran)(ran)(ran)(ran)色。如果使(shi)(shi)用Goldview或(huo)EB等染(ran)(ran)(ran)(ran)(ran)料進行染(ran)(ran)(ran)(ran)(ran)色,由于此(ci)類染(ran)(ran)(ran)(ran)(ran)料帶更多的(de)正電(dian)(dian)荷(he)(染(ran)(ran)(ran)(ran)(ran)料移動方向(xiang)與核酸泳動方向(xiang)相反),隨著電(dian)(dian)泳時(shi)間延長,染(ran)(ran)(ran)(ran)(ran)料向(xiang)大片(pian)段聚集,凝膠(jiao)會(hui)出現陰陽膠(jiao)現象(染(ran)(ran)(ran)(ran)(ran)料含(han)量高的(de)凝膠(jiao)紫(zi)外下(xia)較亮(liang),含(han)量低的(de)凝膠(jiao)紫(zi)外下(xia)發暗),導致小(xiao)分子量DNA顯色很弱或(huo)不可見。陰陽膠(jiao)可以在電(dian)(dian)泳結束(shu)后(hou)浸泡(pao)染(ran)(ran)(ran)(ran)(ran)色,將膠(jiao)浸沒在含(han)1 μg /ml EB或(huo)Goldview的(de)電(dian)(dian)泳緩沖液中20-25 min,小(xiao)分子量DNA會(hui)顯色清楚(chu)。更長時(shi)間浸泡(pao)會(hui)因DNA分子擴散而使(shi)(shi)條帶彌散,小(xiao)分子量DNA更易(yi)彌散。

5. 紫(zi)外照射(she)下EB易分解(jie),使DNA條帶(dai)熒光(guang)變淺。DNA受(shou)紫(zi)外光(guang)照射(she)形成嘧啶(ding)二聚(ju)體,不利于待回(hui)收片段的下游工作,因(yin)此盡可能減少紫(zi)外照射(she)時(shi)間(jian)。

6. 蔗糖會結合DNA,降低(di)其電泳遷移(yi)率(lv),建議DNA樣(yang)品上樣(yang)使用含甘油的Loading Buffer。

7. 大部分核酸工具酶會結合DNA,降低其電泳(yong)遷(qian)移率(lv),大分子(zi)量DNA尤為明顯。
