彩虹預染(ran)超低分子量(liang)蛋白質Marker可以直接觀察蛋白電泳(yong)及清晰(xi)判(pan)斷Western Blotting的(de)轉(zhuan)膜(mo)效果。本(ben)產品包含3種多肽和3種低分(fen)子量蛋白(bai)質組成,分(fen)子量范圍為1.2kD-45kD,分子量大小為 1.2kD,4.6kD,10kD,16kD,27kD,45kD (注:蛋白(bai)大(da)小已在18%Tricine-SDS-PAGE中(zhong)用非預染(ran)蛋白Marker標定過。
● 使用說明(ming):
第一次收到該產品,常溫融化后,徹底混勻,離心快甩將溶液完全收集到管底。
一. 制膠:
I 配制分離膠
1.按照表一將不同體積的雙蒸水、40% PAA(19:1)、4×凝膠緩沖液(貨號:GB010)和乙二醇加入(ru)到(dao)小燒杯(bei)中混合。
2.加入10%APS和TEMED,立即混勻5-10秒,以使溶液充分(fen)混勻。
3.在凝膠模具中迅速灌入適量分離膠溶液,然后在分離膠溶液上輕輕覆蓋一層1
cm的無(wu)水乙(yi)醇,使凝膠(jiao)表面保持平整。
4.靜置15-30分鐘(zhong),待(dai)分離膠和乙(yi)醇(chun)層之間出現一個清晰(xi)的界面表(biao)示(shi)凝膠已聚合(he)。

II 配制濃縮膠
去除覆蓋(gai)在分(fen)離膠上的乙醇(chun)層,用濾紙將殘(can)留乙醇(chun)吸去。
1.按照表一將不(bu)同體積的雙蒸(zheng)水(shui)、40%PAA(29:1)和4×凝(ning)膠緩(huan)沖液加(jia)入到(dao)小燒杯中混合。
2.加入10%過(guo)硫酸(suan)銨(an)和TEMED,立即混勻5-10秒,以使溶液(ye)充分(fen)混勻。
3.將(jiang)梳子插入凝膠內(nei),避免產生氣(qi)泡。
4.靜置30-60分鐘裝(zhuang)待(dai)凝膠聚合
二. 電泳:
1. 取Marker樣品,充(chong)分(fen)混(hun)勻(yun)后備(bei)用。不要(yao)加(jia)熱處理。
2. 電(dian)(dian)泳(yong)前,將(jiang)(jiang)10×陽極(ji)緩(huan)沖液(ye)(Cat No:AB080)和10×陰極(ji)緩(huan)沖液(ye)(Cat No:CB010)用(yong)蒸餾(liu)水(shui)稀釋成1×緩(huan)沖液(ye)備用(yong)。將(jiang)(jiang)電(dian)(dian)泳(yong)槽(cao)的外槽(cao)加(jia)入(ru)(ru)1×陽極(ji)緩(huan)沖液(ye),內(nei)槽(cao)加(jia)入(ru)(ru)1×陰極(ji)緩(huan)沖液(ye),輕柔(rou)拔出梳子,將(jiang)(jiang)Marker或蛋白樣(yang)品(已(yi)經(jing)過2×Tricine上樣(yang)緩(huan)沖液(ye)[Cat No:TP050]處(chu)理)加(jia)入(ru)(ru)點樣(yang)孔,參(can)考下表(biao)進行電(dian)(dian)泳(yong),至每條帶都(dou)清晰可見時(shi)停止(zhi)電(dian)(dian)泳(yong)。整個電(dian)(dian)泳(yong)過程大約(yue)需要2-3個小時(shi)。

三. 染色
根據常規實驗步驟,用配方6和7(表三)進行染色和觀察。如果使用配方6進行染色時效果不好或考慮其毒性,請選擇本公司的
FastBlue蛋白染色液(貨號:RTD6202),該產品除具有染色快,無毒,靈敏性高等特點外,還能對小肽在染色中起到固定作用,不至于小肽在染色和脫色中從凝膠中脫離,是常規染色液的理想替代品。
